News Prostate Health Index and its Possible Use in Clinical Practice
With the increasing incidence of prostate cancer, the effort to improve its diagnostics is also growing. The exclusively used and practice-proven prostate-specific antigen (PSA) continues to play an indispensable role in identifying at-risk patients, but some of its isoforms can also provide important information for deciding the patient's further fate. Based on their combination, a so-called prostate health index (PHI) can be determined. A recently published work by Czech authors assessed the usability of PHI as a parameter suitable for evaluating the aggressiveness of prostate cancer (PC) within the PHIMOPLUKTHOM study.
News Treatment Options for Advanced Parkinson's Disease Using Medical Devices (DAT)
A new treatment option for advanced Parkinson's disease (PD) using medical devices (DAT − device-aided therapy) is the intestinal infusion gel containing levodopa, entacapone, and carbidopa (LECIG). An article published in 2022 in the journal Therapeutic Advances in Neurological Disorders provides an overview of current DAT modalities in PD patients and presents LECIG as the latest extension of options in this area.

The association between type 2 diabetes (DM2) and atherosclerotic cardiovascular disease (ASCVD) is often referred to as cardiometabolic syndrome, while the relationship between chronic kidney disease (CKD) and heart failure (HF) is known as cardiorenal syndrome. Additionally, an increased prevalence of ASCVD, CKD, and HF is known among patients with type 2 diabetes. A recent study investigated the hypothesis that both syndromes constitute a single entity − the so-called cardio-renal-metabolic syndrome.

Combined analgesics were historically recommended exclusively for the treatment of acute or postoperative pain, mainly due to the risk of individual components which, although potentiated their analgesic effect, also exhibited significant organ toxicity or led to long-term and difficult-to-resolve abuses. In recent years, however, fixed combinations containing fewer components have increasingly appeared on the market, and thanks to the optimal synergy of their effects, they can be used in smaller and safe doses. One such combination is tramadol with paracetamol, the place and possibilities of which in the treatment of chronic pain are discussed in the following text.

Psoriasis, psoriatic arthritis (PsA), and axial spondyloarthritis (SpA) are chronic inflammatory diseases that often require long-term treatment. The current pharmacotherapy of these diseases includes conventional synthetic disease-modifying antirheumatic drugs (DMARDs) and targeted biological therapies that inhibit the effects of tumor necrosis factor (TNF) or interleukins IL-23 and IL-17. An analysis published in early 2020 in the journal Rheumatology focused on the safety of long-term inhibition of IL-17A using ixekizumab.

News Mechanisms Connecting Diabetes Mellitus and Obesity
Obesity has a close relationship with insulin resistance and diabetes mellitus (DM); weight gain is associated with a higher incidence of both type 1 and type 2 DM. In the development of type 2 DM, depletion of pancreatic β-cells plays a crucial role, which can no longer produce sufficient insulin to maintain glucose homeostasis. The development of diabetes is much more likely if β-cell failure is accompanied by increased insulin resistance. In obese patients, there is a higher proportion of unsaturated fatty acids, glycerol, pro-inflammatory cytokines, hormones, and other substances in the blood, which have a fundamental impact on insulin resistance.
