Overview

Aim: The goal of the thesis was to analyze samples of urinary sediment and solid biominerals using microscopic and spectroscopic methods. Material and Methods: Biological samples of urinary sediment were evaluated using microscopic analysis and image analysis evaluation. Furthermore, individual types of solid calculi were identified using advanced methods of FTIR spectroscopic analysis. Results: In 2025, 55,443 urinary sediment examinations were performed at Motol University Hospital using automated microscopic analysis followed by image analysis. The main groups of identified crystals include oxalates (1,952), uric acid (624), phosphates (370), and urates (19). In addition, highly specialized spectroscopic examinations of biomaterials (calculi) are performed (approximately 250–350 per year). Various forms of oxalates predominate in this group. Conclusion: Modern spectroscopic methods allow for minimalistic sample preparation. The acquired data are mathematically evaluated and can serve as a very important basis for determining a diagnosis.
