Influence of industrial mineral fiber dusts on respiration toxicity parameters – time dependence

Overview

The objective of the experimental investigation was to determine lung damage – respiratory toxicity – after exposure to selected industrial fiber dusts (glass fibers – GF, ceramic fibers – CF and amosite asbestos– AV as positive control) from the standpoint of time dependence. Animals were instilled with 4 mg glass fibers in 0,4 ml saline solution/animal, the control group with 0,4 ml saline solution. Wistar rats were sacrificed and bronchoalveolar lavage (BAL) were performed after instillation – 2, 30, 90 days. The authors examined selected inflammation, cytotoxic, immunotoxic, clastogenic, genotoxic parameters, the antioxidant state parameters and lung histology [number of BAL cells and alveolar macrophages (AM); differential picture of BAL cells (% AM, polymorphonuclear cells and lymphocytes), % of immature AM cells, multinuclear cells, viability and phygocytic activity of AM cells, frequency of micronuclei (MN), comet assay, FRA (ferric reducing ability) and histology of the lung tissue]. The influence of the industrial dusts under observation on the changes of lung tissue decreased in the following order: AMO > KV > SV. The course of time dependence curves of BAL parameters after exposure to all industrial fibers under investigation proved to be very similar. The explanation of their different consequences (finally the origin of disease) may be in their different biopersistence.
